Yuki Cross's earliest memory is of a snowy night in winter, when she was attacked by one vampire and was saved by another, Kaname Kuran.Yuki is the adopted daughter of the Headmaster of Cross Academy, Cross Kaien and has grown up and became a prefect for the school with one main purpose: to protect the Human Day Class Students and the Vampire Night Class Students from each other. Her fellow guardian, Zero Kiryu, a childhood companion whose parents were killed by a pureblood vampire, is now determined to kill them all and refuses to trust them. Through a cruel twist of fate, Zero becomes a vampire himself, having been bitten by a pure-blood vampire four years before, Shizuka Hio. Kaname leads the group of Night class as their President, with the help of Takuma Ichijo, the Vice-president.

黑主优姬：是作为枢的妻子而出生的。日间部；黑主学院理事长的养女。遭遇吸血鬼时被枢所救，对枢有好感，喜欢枢 武器：狩猎女神阿尔忒弥斯。学园守护者之一。是枢的妹妹兼未婚妻对于零，希望能够守护着他，不让他受到伤害，但是变成吸血鬼后对于作为猎人的零有很深的愧疚感。TV版里，最后与枢还有其他吸血鬼一起离开了黑主学园 Yūki Cross: The adoptive daughter of the headmaster of Cross Academy, Yuki is a member of the school's disciplinary committee (public morals committee in Japanese), which is the 'front' for the Guardians. 10 years prior to the events of Vampire Knight, Yuki was attacked by a vampire, but was rescued by Kaname. She has lost all memory of her life before the attack, and Headmaster Cross took her in as his own daughter. Yuki has a deep affection for Kaname, but realized the implications of his vampiric nature when she witnessed him drinking another vampire's blood when she was in junior high school. She still likes him, but also has a bond with Zero (she lets him
drink her blood). Yuki has a cheerful and easygoing personality, and thanks to her work as a Guardian she is good at keeping her cool in sticky situations. There has been hints that she is special, given that vampires are especially drawn to her blood. Her weapon of choice is 'Artemis', an extendable staff she received from Headmaster Cross.
锥生零：风纪委员之一。出生于吸血鬼猎人家族，沉默寡言，喜欢优姬，10年前父母被纯血种吸血鬼——绯樱闲杀害，虽然后来黑主理事长收养了他，但是锥生已经被绯樱闲咬伤，成为了LEVEL E吸血鬼，家人的死给他带来了巨大伤痛，自此他对吸血鬼深恶痛绝（特别是纯血种）。他是极少数的对血液药锭产生排斥的麻烦的吸血鬼，吃了之后会吐。所以一直在吸食优姬的血。在零吃了弟弟一缕，体内有三个纯种吸血鬼之血（玖兰优姬，玖兰枢，绯樱闲）的零变成了最强大的吸血鬼猎人。日间部；吸血鬼猎人。武器：血蔷薇之。学园守护者之一
Zero Kiryu: Yuki's childhood friend, Zero is also a Guardian and likewise on the disciplinary committee. Zero's family, a line of vampire hunters, was murdered by a pureblood vampire named Shizuka when he was little. He
and his twin brother were the only survivors. He harbors a deep hatred for vampires, and claims to have taken on the role of Guardian to learn to hunt them better. In truth, because Zero was bitten during the attack on his family, he has become a vampire himself. His weapon is the gun 'Bloody Rose', and he had made Yuki promise that if he becomes a Level E vampire one day, she will kill him with that gun.

Kaname Kuran: A pureblood vampire. He saved Yuki from an attack by another vampire when she was little. Kaname is the class president of the Night Class, much feared and respected by the other Night Class students. While he is cold and authoritative towards his fellows in the Night Class, he is always kind and sweet to Yuki. He has shown that he cares deeply for Yuki ever since he saved her, to what might be love in a romantic sense. He seems to know the pureblood Shizuka Hio (he calls her by her first name), who bit Zero
